Abstract
The invention provides silicate prodrugs comprising a therapeutic agent linked to one or more groups of formula (I): —Si(OR)3 (I); wherein each R independently has any of the values defined herein, as well as nanoparticles comprising such compounds.

A compound comprising a taxane linked to one or more groups of formula (I):
- —Si(OR)3 (I)
- wherein:
- each R is independently selected from (C1-C20)alkanoyl, (C2-C20)alkenylcarbonyl, (C2-C20)alkynylcarbonyl, wherein each (C1-C20)alkanoyl, (C2-C20)alkenylcarbonyl, and (C2-C20)alkynylcarbonyl, is optionally substituted with one or more hydroxy, (C1-C6)alkoxy, oxo, halo, or aryl.
A compound of formula (Ia):
- wherein R1 is a group of formula (I),
- —Si(OR)3 (I)
- wherein:
- each R is independently selected from (C1-C20)alkyl, (C2-C20)alkenyl, (C2-C20)alkynyl, (C1-C20)alkanoyl, (C2-C20)alkenylcarbonyl, and (C2-C20)alkynylcarbonyl, wherein each (C1-C20)alkyl, (C2-C20)alkenyl, (C2-C20)alkynyl, (C1-C20)alkanoyl, (C2-C20)alkenylcarbonyl, and (C2-C20)alkynylcarbonyl, is optionally substituted with one or more hydroxy, (C1-C6)alkoxy, oxo, halo, or aryl;
- or a salt thereof.
