Abstract
The present invention relates to a method for the production of a permanent human cell line, wherein isolated primary human cells are transfected simultaneously with a sequence allowing the expression of at least one cell transforming factor and a sequence allowing the expression of at least one recombinant polypeptide.
Claims21
A method for the production of a permanent human cell line comprising transfecting primary human cells with a nucleic acid molecule or at least two distinct nucleic acid molecules, wherein in case that one nucleic acid molecule is transfected, said nucleic acid molecule exhibits a sequence allowing the expression of at least one cell transforming factor and the expression of at least one recombinant polypeptide, wherein the recombinant polypeptide is a hormone, a clotting factor, a growth factor, the Coxsackie and adenovirus receptor (CAR), a viral, bacterial or parasitic antigen, or complementation factors for the production of recombinant viruses, and wherein in case that at least two distinct nucleic acid molecules are transfected, a first nucleic acid molecule exhibits a sequence allowing the expression of at least one cell transforming factor, and a second nucleic acid molecule exhibits a sequence allowing the expression of at least one recombinant polypeptide, wherein transfecting said first and second nucleic acid molecules occurs simultaneously, and wherein the recombinant polypeptide is a hormone, a clotting factor, a growth factor, the Coxsackie and adenovirus receptor (CAR), a viral, bacterial or parasitic antigen, or complementation factors for the production of recombinant viruses.
