Abstract
Compounds of formula (I) and pharmaceutically acceptable salts thereof: wherein R2, W, A, Y and R1 are as defined in the specification, are p38 MAPK inhibitors, and are useful as anti-inflammatory agents in the treatment of, inter alia, diseases of the respiratory tract.
Claims20
A method of treating a disease or condition in a human subject which benefits from inhibition of p38 MAP kinase activity, comprising administering, to a subject in need thereof, a compound represented by formula (I) or a pharmaceutically acceptable salt thereof:
- wherein;
- W is N substituted with hydrogen;
- Y is O;
- R1 is a group represented by formula (IIc):
- X2 is a —CH— group, X3 is a nitrogen atom, X4 is a carbon atom, and X5 is a nitrogen atom;
- R11 is linked to X4 and is C1-C6 alkyl which is substituted by —ORC or —SRC;
- —NRARB;
- —(C1-C6)alkylene-NRARB;
- or —(C3-C7)heterocycloalkyl, wherein any of said NRARB, —(C1-C6)alkylene-NRARB, or —(C3-C7)heterocycloalkyl may be optionally substituted by one or two C1-C6 alkyl;
- RA and RB form, together with the nitrogen atom to which they are attached, a 5-11-membered saturated heterocyclic monocyclic ring system which is optionally substituted by one or more C1-C6 alkyl and which 5-11-membered saturated heterocyclic monocyclic ring optionally contains a further heteroatom which is oxygen or nitrogen;
- RC is at each occurrence independently hydrogen or C1-C6 alkyl, said C1-C6 alkyl being optionally substituted —ORD;
- RD is at each occurrence independently hydrogen, —CH3, or —C2H5;
- R13 is independently hydrogen, C1-C6 alkyl, or halogen;
- A is a group represented by one of the following formulae:
- and
- R2 is a group of formula (IIIa):
- wherein
- R14, R15, and R16 are —CH3,
- wherein said disease or condition is chronic obstructive pulmonary disease, chronic bronchitis, lung fibrosis, pneumonia, acute respiratory distress syndrome, adult respiratory distress syndrome, emphysema, asthma, cystic fibrosis, pulmonary fibrosis, bronchiectasis, exacerbation of airways hyper-reactivity consequent to other drug therapy, or airways disease that is associated with pulmonary hypertension.
