Abstract
The present invention relates to an improved method for synthesizing meropenem trihydrate[(1R,5S,6S)-2-[((2′S,4′S)-2′-dimethylaminocarbozyl)pyrrolidin-4′-ylthio]-6-[(R)-1-hydroxyethyl]-1-methylcarbapen-2-em-3-carboxylic acid, trihydrate], which is a novel carbapenem antibiotic.
Claims17
A method for preparing meropenem trihydrate of formula (1):
- comprising:
- a. reacting meropenem-PNB of formula (2)
- with zinc powder in a mixture solvent comprised of an organic solvent and an 0.5 to 1.6 M KH2PO4 or NaH2PO4 aqueous solution to remove p-nitrobenzyl from said meropenem-PNB, wherein said mixture solvent does not contain any phosphate other than said potassium phosphate or sodium phosphate;
- b. removing phosphate from the mixture resulting from step a;
- and
- c. crystallizing meropenem trihydrate by the addition of a crystallization solvent.
