Abstract
Methods are described for generating autologous tissue grafts, including generating grafts at the point of care, which include isolated cell populations that are enriched with stem cells and are mixed with hyaluronic acid and derivatives thereof. The hyaluronic acid localizes the cells to a desired injection site and stimulates collagen production thus enhancing the viability and the longevity of the graft.
Claims9
A method of preparing and administering, at a point of care, an injectable cell graft for treating a tissue defect in a mammal comprising:
- isolating an autologous stromal vascular fraction (SVF) cell preparation from an adipose tissue of a mammal by releasing cells from a connective tissue matrix of the adipose tissue and substantially removing debris and adipocytes without subjecting the autologous SVF cell preparation to expansion in culture and without subjecting the autologous SVF cell preparation to plastic adherence;
- mixing the isolated autologous SVF cell preparation with hyaluronic acid (HA) and/or a hyaluronic acid derivative (HAD) at the point of care to generate a HA and/or HAD stabilized autologous SVF graft;
- and
- injecting or implanting the HA and/or HAD stabilized autologous SVF graft into the tissue defect of the mammal at the point of care without culturing of the HA and/or HAD stabilized autologous SVF graft.
A method of preparing and implanting, at a point of care, a hyaluronic acid (HA) and/or a hyaluronic acid derivative (HAD) stabilized cell graft in less than four hours comprising:
- isolating a mixed population of stromal vascular cells from a lipoaspirate of a patient at the point of care by releasing cells from the lipoaspirate and removing connective tissue and debris without culture and without plastic adherence of the mixed population of stromal vascular cells, said cells including multi-potent progenitor cells, endothelial cells, and fibroblasts;
- and
- mixing the isolated mixed population of stromal vascular cells with HA and/or HAD, thereby generating a a HA and/or HAD stabilized cell graft that is implanted into the patient at the point of care in less than four hours.
A method of treating a tissue defect in a mammal comprising:
- isolating, at a point-of-care, an autologous stromal vascular cell population by enzymatic digestion of lipoaspirate and removal of connective tissue and debris, wherein <10% of the isolated cells express markers associated with stemness;
- mixing the isolated autologous stromal vascular cell population with hyaluronic acid (HA) and/or a hyaluronic acid derivative (HAD) to provide a HA and/or HAD stabilized autologous cell graft;
- and injecting the HA or HAD stabilized autologous cell graft into the tissue defect at the point of care without pre-adherence and without culture of the autologous stromal vascular cells.
