Abstract
Methods and materials related to producing 3-HP as well as other organic compounds are disclosed. Specifically, isolated nucleic acids, polypeptides, host cells, and methods and materials for producing 3-HP and other organic compounds are disclosed.
Claims41
A transformed yeast or bacterial cell for producing 3-hydroxypropionic acid (3-HP), wherein the transformed cell comprises a metabolic pathway that produces:
- acetyl-CoA from pyruvate,
- malonyl CoA from the acetyl-CoA, and
- 3-HP from the malonyl CoA,
- wherein the transformed cell comprises at least one exogenous nucleic acid molecule that does not naturally occur in the transformed cell and encodes acetyl-CoA carboxylase, a polypeptide that reduces malonyl-CoA, or combinations thereof.
A transformed yeast or bacterial cell for producing 3-hydroxypropionic acid (3-HP), wherein the transformed cell comprises a metabolic pathway that produces:
- oxaloacetate from phosphoenolpyruvate (PEP) or pyruvate,
- aspartate from the oxaloacetate,
- β-alanine from the aspartate,
- malonate semialdehyde from the β-alanine, and
- 3-HP from the malonate semialdehyde,
- wherein the transformed cell comprises at least one exogenous nucleic acid molecule that that does not naturally occur in the transformed cell and encodes PEP carboxylase, PEP carboxykinase, PEP transcarboxylase, pyruvate carboxylase, aspartate aminotransferase, aspartate alpha decarboxylase, 4-aminobutyrate aminotransferase, 3-HP dehydrogenase, 3-hydroxypropionate dehydrogenase, 3-hydroxyisobutyrate dehydrogenase, or combinations thereof.
