Abstract
Provided herein are polycyclic β-lactam compounds, compositions thereof and methods for the treatment and prevention of diseases such as cancer, including pancreatic cancer and pancreatic cancers resistant to mainline chemotherapeutic agents.

A compound of the formula:
- wherein:
- R1 is alkyl(C≦8), alkenyl(C≦8), alkynyl(C≦8), aryl(C≦8), aralkyl(C≦8), heteroaryl(C≦8) or heteroaralkyl(C≦8), or a substituted version of any of these groups;
- and
- R4 is:
- hydrogen;
- or a pharmaceutically acceptable salt or tautomer thereof.

A method for treating cancer, wherein the cancer is colorectal cancer, breast cancer, pancreatic cancer, lung cancer, skin cancer, or prostate cancer, in a patient in need thereof comprising administering to the patient an effective amount of a compound of the formula:
- wherein:
- R1 is alkyl(C≦8), alkenyl(C≦8), alkynyl(C≦8), aryl(C≦8), aralkyl(C≦8), heteroaryl(C≦8) or heteroaralkyl(C≦8), or a substituted version of any of these groups;
- and
- R4 is hydrogen;
- or a pharmaceutically acceptable salt thereof.
