Abstract
An ocular implant comprising siRNA complexed with a transfection agent selected from the group consisting of cationic lipids and short cell penetration peptides, wherein the complex is associated with a biocompatible polymer is described. The biocompatible polymer comprises a polymeric matrix configured to release the complex into the eye of a patient at therapeutic levels for a time sufficient to treat an ocular condition or disease.
Claims6
A hot-melt extruded, rod-shaped ocular implant comprising a small interfering RNA (siRNA) complexed with a transfection agent, wherein the transfection agent is selected from the group consisting of polylysine peptides, polyarginine peptides, R9, Pep-1 (SEQ ID NO:1), and MPG (SEQ ID NO.:2), wherein said complex is associated with a biodegradable polymeric matrix consisting essentially of a poly(D,L-lactide-co-glycolide) polymer, a poly(D,L-lactide) polymer, or both a poly(D,L-lactide-co-glycolide) copolymer and a poly(D,L-lactide) polymer, wherein said polymeric matrix is configured to release said complex into the eye of a patient at therapeutic levels for a time sufficient to treat an ocular condition or disease.
A hot-melt extruded, rod-shaped biodegradable ocular implant comprising a small interfering RNA (siRNA), a transfection agent, and a biodegradable polymer, wherein the siRNA is complexed with the transfection agent, and wherein said complex is associated with the biodegradable polymer, wherein the transfection agent is selected from the group consisting of a polylysine peptide, a polyarginine peptide, R9, Pep-1 (SEQ ID NO:
- 1), and MPG (SEQ ID NO:
- 2), wherein the polylysine peptide is an amino acid chain of between 3 and 50 contiguous lysines, wherein the polyarginine peptide is an amino acid chain of between 3 and 50 contiguous arginines, and wherein R9 is a polyarginine peptide consisting of 9 contiguous arginines.
