Disclosed herein are methods of preparing a phosphorothioate nucleotide analog, which are useful in treating diseases and/or conditions such as viral infections.

A compound having the structure:
A method of preparing a compound of Formula (B) comprising:
- reacting O-phenylphosphorodichloridothioate with (S)-isopropyl 2-aminopropanote.
A method of preparing a compound of Formula (I):
- wherein the method comprises:
- coupling a compound of Formula (A) and a compound of Formula (B) in a solvent, wherein the —OH groups and —NH group of the compound of Formula (A) are unprotected during the coupling reaction;
- and wherein the solvent is methyl isobutyl ketone.
