Abstract
Provided herein are methods of treating diseases mediated by PIM-1 and/or PIM-2 and/or PIM-3 kinases by administering to a mammal in need thereof a therapeutically effective amount of a compound of Formula I: in which B, R1, R1a, R2, R3, R4, R5, R6, R7, R10 and R11 have the meanings given in the specification.
Claims22
A method of inhibiting PIM-1 and/or PIM-2 and/or PIM-3 in a mammal, which comprises administering to said mammal a therapeutically effective amount of a compound of general Formula I:
- or a pharmaceutically acceptable salt thereof, wherein:
- R10 and R11 together with the N to which they are attached form a 4-8 membered heterocyclic ring optionally having an additional ring heteroatom selected from N and O, wherein the heterocyclic ring is optionally substituted with one or more R9 groups;
- each R9 is independently selected from halogen, (1-6C)alkyl, NRfRg, -(1-6C alkyl)NRhRi, ORj, (1-6C alkyl)ORk, C(O)NRmRn, C(O)O(1-6C alkyl), and -(1-6C alkyl)NRhC(O)O(1-6C alkyl);
- B is H, ORa, (1-6C alkyl)NRbRc, (1-6C alkyl)OH, NRbRc, or CH(OH)CH2OH;
- R1 is H, F, Cl, Br, methyl, ethyl, cyclopropyl or CN;
- R1a, R2, R3 and R4 are independently H, F, Cl, Br, methyl, ethyl, cyclopropyl or CN;
- R5 and R7 are independently H, F, Me or CN;
- R6 is H, F, Me, Br, CN, cyclopropyl or phenyl;
- Ra is H, (1-6C alkyl), -(1-6C alkyl)-O-(1-6C alkyl) or -(1-6C alkyl)-O-(3-6C cycloalkyl);
- each Rb, Rc, Rd, Re, Rf, Rg, Rh, Ri, Rj, Rk, and Rm is independently selected from H and (1-6C alkyl);
- and
- Rn is H, (1-6C alkyl) or O-(1-6C alkyl).
A method of treating a cancer that overexpresses one or more PIM kinases selected from PIM-1 and/or PIM-2 and/or PIM-3 in a mammal, which comprises administering to said mammal a therapeutically effective amount of a compound of general Formula I:
- or a pharmaceutically acceptable salt thereof, wherein:
- R10 and R11 together with the N to which they are attached form a 4-8 membered heterocyclic ring optionally having an additional ring heteroatom selected from N and O, wherein the heterocyclic ring is optionally substituted with one or more R9 groups;
- each R9 is independently selected from halogen, (1-6C)alkyl, NRfRg, -(1-6C alkyl)NRhRi, ORj, (1-6C alkyl)ORk, C(O)NRmRn, C(O)O(1-6C alkyl), and -(1-6C alkyl)NRhC(O)O(1-6C alkyl);
- B is H, ORa, (1-6C alkyl)NRbRc, (1-6C alkyl)OH, NRbRC, or CH(OH)CH2OH;
- R1 is H, F, Cl, Br, methyl, ethyl, cyclopropyl or CN;
- R1a, R2, R3 and R4 are independently H, F, Cl, Br, methyl, ethyl, cyclopropyl or CN;
- R5 and R7 are independently H, F, Me or CN;
- R6 is H, F, Me, Br, CN, cyclopropyl or phenyl;
- Ra is H, (1-6C alkyl), -(1-6C alkyl)-O-(1-6C alkyl) or -(1-6C alkyl)-O-(3-6C cycloalkyl);
- each Rb, Rc, Rd, Re, Rf, Rg, Rh, Ri, Rk, and Rm is independently selected from H and (1-6C alkyl);
- and
- Rn is H, (1-6C alkyl) or O-(1-6C alkyl).
