Abstract
This invention relates to attenuated pestiviruses characterized in that their enzymatic activity residing in glycoprotein ERNS is inactivated, as well as methods of preparing, using and detecting these pestiviruses.
Claims6
A method for attenuating a CSFV pestivirus comprising inactivating the RNase activity of glycoprotein ERNS in said pestivirus, wherein the RNase activity is inactivated by the substitution of the histidine residue located in the conserved ERNS sequence SLHGIWPEKIC (SEQ ID NO:7) with a leucine.
A method for producing an immunogenic composition comprising a CSFV pestivirus comprising the steps of:
- a) attenuating said pestivirus by inactivating the RNase activity of glycoprotein ERNS in said pestivirus, wherein the RNase activity is inactivated by the substitution of the histidine residue located in the conserved ERNS sequence SLHGIWPEKIC (SEQ ID NO:7) with a leucine;
- and
- b) admixing a pharmaceutically acceptable stabilizer/carrier with said attenuated pestivirus;
- wherein said composition induces an immunological response in a vaccinated animal.
A method for attenuating a CSFV pestivirus comprising inactivating the RNase activity of glycoprotein ERNS in said pestivirus by the deletion of the second histidine residue located in the conserved ERNS sequence RHEWNKHGWCNW (SEQ ID NO:
- 8).
A method for producing an immunogenic composition comprising a CSFV pestivirus comprising the steps of:
- (1) attenuating said pestivirus by inactivating the RNase activity of glycoprotein ERNS in said pestivirus by the deletion of the second histidine residue located in the conserved ERNS sequence RHEWNKHGWCNW (SEQ ID NO:8);
- and
- (2) admixing a pharmaceutically acceptable stabilizer or carrier, wherein said composition induces an immunological response in a vaccinated animal.
A method for attenuating a BVDV pestivirus comprising inactivating the RNase activity of glycoprotein ERNS in said pestivirus by the deletion of the second histidine residue located in the conserved ERNS sequence RHEWNKHGWCNW (SEQ ID NO:8).
A method for producing an immunogenic composition comprising a BVDV pestivirus comprising the steps of:
- (1) attenuating said pestivirus by inactivating the RNase activity of glycoprotein EP′Ns in said pestivirus by the deletion of the second histidine residue located in the conserved ERNs sequence RHEWNKHGWCNW (SEQ ID NO:8);
- and
- (2) admixing a pharmaceutically acceptable stabilizer or carrier, wherein said composition induces an immunological response in a vaccinated animal.
