Abstract
Lentiviral vectors modified at the 5′ LTR or both the 5′ and 3′ LTR's are useful in the production of recombinant lentivirus vectors. Such vectors can be produced in the absence of a functional tat gene. Multiple transformation of the host cell with the vector carrying the transgene enhances virus production.
Claims18
A lentivirus packaging plasmid wherein endogenous sequence upstream from gag gene is deleted, wherein nef gene and all endogenous sequence downstream from env gene are deleted, wherein the endogenous sequence upstream from gag gene is selected from the group consisting of all endogenous sequence upstream from gag gene and all endogenous sequence upstream from gag gene except for the 5′ splice donor site, wherein the endogenous sequence upstream from the gag gene is replaced with a heterologous sequence comprising a heterologous promoter, and wherein said plasmid carries an expressed gag gene, an expressed pol gene or expressed gag and pol genes.
