Abstract
The invention relates to sesquiterpene synthases and methods for their production and use. Particularly, the invention provides nucleic acids comprising the nucleotide sequence of citrus valencene synthase (CVS) which codes for at least one CVS. The invention further provides nucleic acids comprising the nucleotide sequence coding for amino acid residues forming the tier 1 and tier 2 domains of CVS. The invention also provides for methods of making and using the nucleic acids and amino acids of the current invention.
Claims21
A method of producing valencene, comprising:
- providing a host cell comprising a farnesyl pyrophosphate substrate and a nucleic acid encoding a valencene synthase polypeptide, wherein:
- the nucleic acid encoding the valencene synthase polypeptide is heterologous to the cell;
- and
- the nucleic acid encodes a polypeptide that comprises the sequence of amino acid residues set forth in SEQ ID NO:
- 4;
- and
- culturing the host cell under conditions in which the valencene synthase polypeptide encoded by the nucleic acid is expressed, whereby the polypeptide reacts with farnesyl pyrophosphate substrate to catalyze the production of valencene.
A nucleic acid molecule, comprising the sequence of nucleotides set forth in SEQ ID NO:
- 1, wherein the nucleic acid molecule is a cDNA molecule.
A nucleic acid molecule that encodes the polypeptide comprising the sequence of amino acids set forth in SEQ ID NO:
- 4, wherein the nucleic acid molecule is a cDNA molecule.
