Abstract
The present invention relates to compositions comprising complexes of human cells and polymer fibers and methods of their use for therapeutic purposes. Methods of making such compositions are also provided. The present invention encompasses compositions comprising poly-β-1→4-N-acetylglucosamine polymers and stored platelets and their use for promoting wound healing and achieving hemostasis.
Claims44
A method for activating platelets isolated from a mammal for later therapeutic use, the method comprising contacting ex vivo (i) platelets that have expired for transfusion purposes, (ii) poly-β-1→4-N-acetylglucosamine fibers, wherein the poly-β-1→4-N-acetylglucosamine fibers are about 10 to 500 nm in width and about 50 to 750 microns in length as detected by scanning electron microscopy, and (iii) a calcium salt or a calcium salt solution, such that a gel is formed.
The method of claim 1, wherein the platelets activated using the method of claim 12 are activated as determined by a hypotonic stress response assay, a collagen-induced aggregation test, an adenosine diphosphate-induced aggregation test, morphological observation, detection of surface exposure of alpha granule contents, or detection of the conformation of alpha2b beta3 integrin.
A method for activating platelets isolated from a mammal for later therapeutic use, the method comprising contacting ex vivo (i) platelets, and (ii) poly-β-1→4-N-acetylglucosamine fibers, wherein the poly-β-1→4-N-acetylglucosamine fibers are about 10 to 500 nm in width and about 50 to 750 microns in length as detected by scanning electron microscopy, such that a platelet-poly-β-1→4-N-acetylglucosamine composition comprises at least 1×106 of the platelets bound per mg of the poly-β-1→4-N-acetylglucosamine fibers is formed.
