Abstract
The present disclosure describes tetrahydropyran nucleoside analogs, oligomeric compounds prepared therefrom and methods of using the oligomeric compounds. More particularly, tetrahydropyran nucleoside analogs are provided, having one or more chiral substituents, that are useful for enhancing properties of oligomeric compounds including nuclease resistance and binding affinity. In some embodiments, the oligomeric compounds provided herein hybridize to a portion of a target RNA resulting in loss of normal function of the target RNA.
Claims23
An oligomeric compound having from 8 to 40 linked monomeric subunits comprising:
- at least one tetrahydropyran nucleoside analog having one of Formulas Ib or Ic:
- wherein independently for each tetrahydropyran nucleoside analog of said formula:
- Bx is a heterocyclic base moiety;
- X is O or S;
- T6 is H, a hydroxyl protecting group, a 5′ or 3′-terminal group or an internucleoside linking group linking the tetrahydropyran nucleoside analog to at least one of said monomer subunits;
- T7 is at least one of said monomer subunits;
- X is O or S;
- and
- at least one modified nucleoside wherein each of said at least one modified nucleoside is, independently, selected from a bicyclic nucleoside, a 2′-modified nucleoside, a 4′-thio modified nucleoside and a 4′-thio-2′-modified nucleoside;
- wherein said oligomeric compound is hybridizable to at least a portion of a target nucleic acid.
