Abstract
The present invention relates to the isolation and to the characterization of two proteins having a novel enzymatic activity, i.e. a porphyranase activity. These proteins are useful for hydrolyzing polysaccharides containing sulfated agaro-colloids and for producing oligo-porphyrans, notably oligo-porphyrans with a defined structure and size.
Claims8
An isolated polypeptide having porphyranase activity and comprising an amino acid sequence having at least 90% amino acid sequence identity to the sequence of residues 19 to 274 of the amino acid sequence of SEQ ID NO:
- 2 or comprising an amino acid sequence having at least 90% amino acid sequence identity to the sequence of residues 22 to 293 of the amino acid sequence of SEQ ID NO:
- 5.
A method for hydrolyzing polysaccharides comprising the following steps:
- a) providing an isolated polypeptide having porphyranase activity and comprising an amino acid sequence having at least 90% amino acid sequence identity to the sequence of residues 19 to 274 of the amino acid sequence of SEQ ID NO:
- 2 or comprising an amino acid sequence having at least 90% amino acid sequence identity to the sequence of residues 22 to 293 of the amino acid sequence of SEQ ID NO:
- 5, or
- providing an isolated host cell transformed by an expression vector comprising a nucleic acid encoding a polypeptide having porphyranase activity and comprising an amino acid sequence having at least 90% amino acid sequence identity to the sequence of residues 19 to 274 of the amino acid sequence of SEQ ID NO:
- 2 or comprising an amino acid sequence having at least 90% amino acid sequence identity to the sequence of residues 22 to 293 of the amino acid sequence of SEQ ID NO:
- 5;
- and
- b) putting said polypeptide or said host cell in contact with a polysaccharide under conditions for hydrolyzing said polysaccharide to obtain hydrolyzed polysaccharides.
