Abstract
Process, use, method and formulation of disoxyribonucleic acid vaccine inclusion (DNA) in nutritional compositions for animal in culture, particularly in fishery systems.
Claims16
A DNA vaccine for fish, incorporated into a fish food comprising:
- a. a plasmid construct pcDNA-VP2 comprising a 724 bp insert as set forth in SEQ ID NO:
- 5;
- b. a DOTAP Liposome;
- and
- c. a nutritional food base.
2 more elements
A process for obtaining a DNA vaccine for fish, incorporated into fish food, comprising:
- a. constructing a plasmid pcDNA-VP2 comprising SEQ ID NO:
- 5;
- b. transforming a suitable stock host cell with pcDNA-VP2;
- c. amplifying and purifying the pcDNA-VP2 from the stock host cell;
- d. forming a lipoplex that is a binary complex comprising pcDNA-VP2 and a DOTAP liposome, thereby generating a lipoplex;
- e. dispersing, mixing, and integrating the lipoplex into a nutritional food base to make an oral DNA vaccine or a formulated DNA vaccine;
- and
- f. packaging the oral or formulated DNA vaccine.
5 more elements
A pharmaceutical composition comprising a DNA fish vaccine comprising:
- (a) a plasmid comprising the sequence of SEQ ID NO:
- 5;
- (b) a cationic DOTAP liposome as a vehicle;
- and
- (c) nutritional, innocuous, and pharmaceutically acceptable excipients, wherein the excipients are nutritional and digestible for fish.
