Abstract
The present invention refers to agents for modulating the activity of proteins having a PWWP domain.
Claims26
A single-stranded oligonucleotide molecule comprising the sequence
- wherein the sequence
- is 5′-(X1)m-TAC AAC ACC CAC AAA-B-TTT GTG GGT GTT GTA-(X2)m-3′ (SEQ ID NO:1), wherein A, C, T and G represent nucleotide or nucleotide analogue building blocks, X1 and X2 are independently selected from moieties comprising poly(alkylene glycol) units, n and m independently represent 0 or 1, wherein at least one of n and m is 1, and B represents a bridging moiety.
A method for diagnosing or treating conditions or disorders associated with, accompanied by and/or caused by a PWWP domain protein dysfunction, comprising administering a single-stranded oligonucleotide molecule for a PWWP domain protein, to a patient in need of such diagnosis or treatment, wherein said single-stranded oligonucleotide molecule comprises the sequence 5′-(X1)m-TAC AAC ACC CAC AAA-B-TTT GTG GGT GTT GTA-(X2)m-3′ (SEQ ID NO:1), wherein A, C, T and G represent nucleotide or nucleotide, analogue building blocks, X1 and X2 are independently selected from moieties comprising poly(alkylene glycol) units, n and m independently represent 0 or 1, wherein at least one of n and m is 1, and B represents a bridging moiety.
A method for screening for molecules which modulate the interaction of PWWP domain protein oligonucleotide molecules and their target proteins, comprising combining a molecule to be tested with a single-stranded oligonucleotide molecule comprising the sequence 5′-(X1)m-TAC AAC ACC CAC AAA-B-TTT GTG GGT GTT GTA-(X2)m-3′ (SEQ ID NO:1), wherein A, C, T and G represent nucleotide or nucleotide analogue building blocks, X1 and X2 are independently selected from moieties comprising poly(alkylene glycol) units, n and m independently represent 0 or 1, wherein at least one of n and m is 1, and B represents a bridging moiety, and a target protein, and determining whether the molecule to be tested modulates the interaction of the PWWP domain protein oligonucleotide molecule and the target protein.
