Abstract
This invention is metabolically engineer bacterial strains that provide increased intracellular NADPH availability for the purpose of increasing the yield and productivity of NADPH-dependent compounds. In the invention, native NAD-dependent GAPDH is replaced with NADP-dependent GAPDH plus overexpressed NADK. Uses for the bacteria are also provided.
Claims9
A recombinant bacteria comprising i) a disrupted native NAD-dependent D-glyceraldehyde-3-phosphate dehydrogenase (GAPDH), ii) an overexpressed NADP-dependent GAPDH, and iii) an overexpressed NAD kinase, wherein said bacteria has a disrupted gapA gene, plus an overexpressed gapB or gapC gene, and an overexpressed nadK gene.
A recombinant E. coli, comprising i) a deleted native gapA gene, plus one or more expression vectors comprising ii) an overexpressed NADP-dependent GAPDH, and iii) an overexpressed NAD kinase.
