Abstract
The present invention relates to optical confinements, methods of preparing and methods of using them for analyzing molecules and/or monitoring chemical reactions. The apparatus and methods embodied in the present invention are particularly useful for high-throughput and low-cost single-molecular analysis.
Claims25
A method of characterizing a single nucleic acid molecule, comprising:
- a) providing the single nucleic acid molecule, wherein the single nucleic acid molecule comprises a first sequence of contiguous nucleotides;
- b) performing iterative sequencing of the first sequence of contiguous nucleotides in the single nucleic acid molecule by subjecting the single nucleic acid molecule to a template-directed polymerization reaction, wherein the iterative sequencing comprises sequencing the first sequence of contiguous nucleotides in the single nucleic acid molecule multiple times during the polymerization reaction, thereby generating multiple reads of the first sequence of contiguous nucleotides;
- and
- c) analyzing the multiple reads to determine a sequential base composition for the first sequence of contiguous nucleotides, thereby characterizing the single nucleic acid molecule.
