Abstract
The invention relates generally to compositions and methods for altering the isoelectric point of an antibody, and in some cases, resulting in improved plasma pharmacokinetics, e.g. increased serum half-life in vivo.
Claims6
An antibody comprising:
- a) a variant heavy chain constant domain polypeptide comprising a variant of SEQ ID NO:2 wherein said variant comprises the amino acid substitutions S119E/K133E/T164E/K205E/N208D/K210E;
- and
- b) a variant light chain constant domain polypeptide comprising SEQ ID NO:8;
- wherein numbering is according to the EU index as in Kabat.

An antibody comprising:
- a) a variant heavy chain constant domain polypeptide comprising a variant of SEQ ID NO:2 wherein said variant comprises the amino acid substitutions S119E/K133E/T164E/K205E/N208D/K210E;
- and
- b) a variant light chain constant domain polypeptide comprising SEQ ID NO:8;
- wherein numbering is according to the EU index as in Kabat, and wherein said antibody has an increased serum half life in a mammal as compared to an antibody comprising a heavy chain constant domain having SEQ ID NO:2 and a light chain constant domain having SEQ ID NO 1.
