Abstract
Described herein are microorganisms that produce methionine and related products from endogenous genes in a transsulfuration pathway, as well as from exogenous genes providing a direct sulfhydrylation pathway. Novel genes that are useful for methionine and SAMe production are disclosed.
Claims12
An isolated E. coli for producing a detectable amount of methionine, the isolated E. coli comprising:
- one or more first polypeptides comprising direct sulfhydrylation activity, wherein the one or more first polypeptides uses O-succinylhomoserine (OSHS) or O-acetylhomoserine (OAHS) as a substrate and is encoded by metZ, metY or a combination thereof;
- one or more second polypeptides comprising transsulfuration activity, wherein the one or more second polypeptides is encoded by endogenous E. coli metB, metC or a combination thereof;
- and
- a third polypeptide comprising homoserine O-succinyltransferase activity, wherein the third polypeptide is encoded by feedback inhibition resistant metA comprising the amino acid sequence of SEQ ID NO:
- 2, 4, 6, 8, or 10,
- wherein the direct sulfhydrylation activity arises from expression of an exogenous nucleic acid sequence of metZ, metY, or a combination thereof, and
- wherein metZ is obtained from Pseudomonas aeruginosa, and metY is obtained from Leptospira meyeri, Deinococcus radiodurans, Chloroflexus aurantiacus, or Pseudomonas aeruginosa.
