Abstract
This disclosure relates to novel morphinan compounds and their derivatives, pharmaceutically acceptable salts, solvates, and hydrates thereof. This disclosure also provides compositions comprising a compound of this disclosure and the use of such compositions in methods of treating diseases and conditions that are beneficially treated by administering a σ1 receptor agonist that also has NMDA antagonist activity.

A method for the treatment of a subject suffering from or susceptible to a disease or condition selected from emotional lability;
- pseudobulbar affect;
- chronic pain;
- intractable pain;
- neuropathic pain;
- sexual dysfunction;
- intractable coughing;
- dermatitis;
- and Rett syndrome (RTT), comprising administering to the subject a compound according to formula I:
- or a pharmaceutically acceptable salt thereof, wherein:
- R1 is CD3;
- and
- R2 is CH3.

Separate dosage forms of:
- (i) a compound of formula I:
- or a pharmaceutically acceptable salt thereof, wherein:
- R1 is CD3, and
- R2 is CH3;
- or a composition comprising a compound of formula I or pharmaceutically acceptable salt thereof formulated for pharmaceutical administration and a pharmaceutically acceptable carrier;
- and
- (ii) a second therapeutic agent selected from quinidine and quinidine sulfate, wherein said compound, pharmaceutically acceptable salt thereof or composition and second therapeutic agent are associated with one another.
