## Abstract

Materials and methods are provided for producing patterned multi-array, multi-specific surfaces for use in diagnostics. The invention provides for electrochemiluminescence methods for detecting or measuring an analyte of interest. It also provides for novel electrodes for ECL assays. Materials and methods are provided for the chemical and/or physical control of conducting domains and reagent deposition for use multiply specific testing procedures.

## Claims

### Claim 1

An instrument for detection or quantification of an analyte by electrochemiluminescence over a broad range of concentrations, comprising:
(a) a support including a plurality of electrodes each comprising a composite containing a matrix and a multiplicity of carbon particles dispersed therein, wherein said plurality of electrodes includes at least two discrete binding domains wherein
(i) each of said at least two discrete binding domains comprises one or more binding reagents affixed to said electrode, said binding reagents being capable of binding a component of a binding electrochemiluminescence assay; and
(ii) said binding reagents in each of said discrete binding domains recognize said analyte but have different binding affinity for said analyte;
(b) a voltage source to supply an electric potential to said plurality of electrodes;
(c) one or more light detectors capable of detecting or quantifying light emitted from each of said at least two discrete binding domains on said electrodes; and
(d) a microprocessor for detecting or quantifying said analyte based on information received from said one or more light detectors.

### Claim 2 (depends on 1)

The instrument system of claim 1 further comprising a dry reagent on said support.

### Claim 3 (depends on 1)

The instrument system of claim 1, wherein each of said plurality of electrodes is individually addressable.

### Claim 4 (depends on 2)

The instrument of claim 2, wherein said dry reagent is reconstituted during the assay.

### Claim 5 (depends on 1)

The instrument of claim 1, wherein said voltage source is selected from the group consisting of a single voltage source, multiple voltage sources, a computer controlled voltage source, a voltage waveform generator, and combinations thereof.

### Claim 6 (depends on 5)

The instrument of claim 5, wherein the voltage source is a voltage waveform generator.

### Claim 7 (depends on 6)

The instrument of claim 6, wherein the voltage waveform generator is a potentiostat.

### Claim 8

An instrument for detection or quantification of an analyte by electrochemiluminescence over a broad range of concentrations from 10−12 M to 10−3 M, comprising:
(a) a support including a dry reagent thereon, and a plurality of electrodes each comprising a composite containing a matrix and a multiplicity of carbon particles dispersed therein, wherein each of said plurality of electrodes
(i) comprises at least two discrete binding domains,
(ii) comprises one or more binding reagents affixed to said electrode, said binding reagents being capable of binding a component of a binding electrochemiluminescence assay, wherein said binding reagents in each of said discrete binding domains recognize said analyte but have different binding affinity for said analyte, and
(iii) is individually addressable;
(b) a voltage source to supply an electric potential to said plurality of electrodes;
(c) one or more light detectors capable of detecting or quantifying light emitted from each of said at least two discrete binding domains on said electrodes; and
(d) a microprocessor for detecting or quantifying said analyte based on information received from said one or more light detectors.
