Novel intermediates, process for their preparation and their use in the preparation of therapeutically effective antineoplastic agents, in particular Temsirolimus of formula (I).

A process for the preparation of Temsirolimus of formula (I),
- the method comprising converting compound of formula (II),
- wherein R1, R2 and R3 each may be different or same and selected from the group comprising hydrogen, alkyl of C1-C4 carbon atoms, and alkoxy of C1-C4 carbon atoms, to Temsirolimus;
- in the presence of an acid, selected from the group comprising sulfuric acid, hydrochloric acid and mixtures thereof;
- and
- in the presence of a solvent selected from the group comprising alcohols and ethers or mixture thereof.
