Substituted tricyclic compounds and methods of use thereof

A compound of formula (I)

• wherein
• A is N or C—CN,
• D is absent or is —CH2— or —CH(CH3)—,
• E is 0, S or N—R12, wherein
• R12 is hydrogen or (C1-C4)-alkyl,
• L represents —C(═O)—, —S(═O)q— or —S(═O)(═N—R13)—, wherein
• q is 1 or 2,
• and
• R13 is hydrogen or (C1-C4)-alkyl,
• m is 1 or 2,
• n is 1, 2 or 3,
• p is 0, 1 or 2,
• R1 represents hydrogen or halogen,
• R2 represents hydrogen, halogen or (C1-C4)-alkyl,
• R3 is selected from the group consisting of hydrogen, halogen, cyano, (C1-C4)-alkyl, (C1-C4)-alkoxy, (C2-C4)-alkenyl and (C2-C4)-alkinyl,
• R4 is selected from the group consisting of hydrogen, halogen, cyano, (C1-C4)-alkyl, hydroxy, (C1-C4)-alkoxy, (C2-C4)-alkenyl and (C2-C4)-alkinyl,
• R5 is selected from the group consisting of hydrogen, halogen, cyano, (C1-C4)-alkyl, (C2-C4)-alkenyl and (C2-C4)-alkinyl,
• or
• R5 is a group of the formula ♦-O—(CH2)r—R14, wherein
• ♦ denotes the point of attachment,
• r is 0, 1 or 2, and
• R14 represents phenyl or 5- or 6-membered heteroaryl which are optionally substituted with one to three substituents independently selected from the group consisting of halogen, (C1-C4)-alkyl and (C1-C4)-alkoxy,
• or
• R4 and R5 are linked together and form a fused heterocyclic moiety of the formula
• * indicates the point of attachment to the phenyl ring in R4 position,
• ** indicates the point of attachment to the phenyl ring in R5 position,
• and
• R15 represents phenyl or 5- or 6-membered heteroaryl which are optionally substituted with one to three substituents independently selected from the group consisting of halogen, (C1-C4)-alkyl and (C1-C4)-alkoxy,
• R6 represents a substituent independently selected from the group consisting of (C1-C4)-alkyl, hydroxy and (C1-C4)-alkoxy,
• R7 and R8 both represent hydrogen or are taken together to form a bond, resulting in an acetylenic linkage,
• and either
• R9 and R10 are joined, and taken together with the atoms to which they are attached, form a 5- to 7-membered heterocycloalkyl ring which is optionally substituted with one or two substituents independently selected from the group consisting of (C1-C4)-alkyl, hydroxy and (C1-C4)-alkoxy,
• and
• R11 represents hydrogen or (C1-C4)-alkyl,
• or
• R9 is hydrogen,
• and then
• R10 and R11 are independently selected from the group consisting of hydrogen, (C1-C6)-alkyl, (C3-C10)-cycloalkyl and 4- to 7-membered heterocycloalkyl, wherein
• (i) said (C1-C6)-alkyl is optionally substituted with one to three substituents independently selected from the group consisting of fluoro, cyano, hydroxy, (C1-C4)-alkoxy, oxo, amino, mono-(C1-C4)-alkylamino, di-(C1-C4)-alkylamino, (C1-C4)-alkylcarbonylamino, (C1-C4)-alkoxycarbonylamino, hydroxycarbonyl, (C1-C4)-alkoxycarbonyl, aminocarbonyl, mono-(C1-C4)-alkylaminocarbonyl, di-(C1-C4)-alkylaminocarbonyl, (C1-C4)-alkylsulfonyl, S—(C1-C4)-alkylsulfonimidoyl, (C3-C7)-cycloalkyl, phenyl, 5- or 6-membered heteroaryl and 4- to 7-membered heterocycloalkyl,
• wherein said (C3-C7)-cycloalkyl, phenyl, 5- or 6-membered heteroaryl and 4- to 7-membered heterocycloalkyl substituents in turn are optionally substituted with one or two residues independently selected from the group consisting of fluoro, chloro, trifluoromethyl, (C1-C4)-alkyl, hydroxy, (C1-C4)-alkoxy, amino, mono-(C1-C4)-alkylamino and di-(C1-C4)-alkylamino,
• and
• (ii) said (C3-C10)-cycloalkyl and 4- to 7-membered heterocycloalkyl are optionally substituted with one to three substituents independently selected from the group consisting of (C1-C4)-alkyl, oxo, hydroxy, (C1-C4)-alkoxy, amino, mono-(C1-C4)-alkylamino and di-(C1-C4)-alkylamino,
• or
• R10 and R11 are joined, and taken together with the nitrogen atom to which they are attached, form a 3- to 10-membered heterocycloalkyl ring which is optionally substituted with one or two substituents independently selected from the group consisting of (C1-C6)-alkyl, hydroxy, (C1-C4)-alkoxy, amino, mono-(C1-C4)-alkylamino, di-(C1-C4)-alkylamino, (C1-C4)-alkylcarbonylamino, (C1-C4)-alkylcarbonyl, (C1-C4)-alkylsulfonyl, hydroxycarbonyl, (C1-C4)-alkoxycarbonyl, aminocarbonyl, mono-(C1-C4)-alkylaminocarbonyl, di-(C1-C4)-alkylaminocarbonyl, (C3-C7)-cycloalkyl, phenyl, phenoxy, 5- or 6-membered heteroaryl and 4- to 7-membered heterocycloalkyl, wherein
• (i) said (C1-C6)-alkyl substituent in turn is optionally substituted with one or two residues independently selected from the group consisting of hydroxy, (C1-C4)-alkoxy, oxo, amino, mono-(C1-C4)-alkylamino, di-(C1-C4)-alkylamino, (C1-C4)-alkylcarbonyl-amino, (C1-C4)-alkoxycarbonylamino, hydroxycarbonyl, (C1-C4)-alkoxycarbonyl, amino-carbonyl, mono-(C1-C4)-alkylaminocarbonyl, di-(C1-C4)-alkylaminocarbonyl and 4- to 7-membered heterocycloalkyl,
• wherein said (C1-C4)-alkoxy residue is optionally substituted with hydroxy or (C1-C4)-alkoxy,
• and
• (ii) said (C3-C7)-cycloalkyl, phenyl, phenoxy, 5- or 6-membered heteroaryl and 4- to 7-membered heterocycloalkyl substituents in turn are optionally substituted with one or two residues independently selected from the group consisting of fluoro, chloro, tri-fluoromethyl, (C1-C4)-alkyl, hydroxy, (C1-C4)-alkoxy, amino, mono-(C1-C4)-alkylamino and di-(C1-C4)-alkylamino,
• or
• R10 and R11 are joined, and taken together with the nitrogen atom to which they are attached, form a heterocyclic group of the formula
• # denotes the point of attachment to the CHR9 group,
• and
• R16 represents hydrogen, trifluoromethyl or (C1-C4)-alkyl,
• or a pharmaceutically acceptable salt thereof.
