A floating separating element for use in centrifugal separation of components of a physiological fluid comprises a positioning part and a separating part, where the positioning part is designed to automatically assume a position in a supernatant and a separating part is positioned at a desired location with respect to the interface between the supernatant and heavier components. In preferred embodiments the physiological fluids are blood or bone marrow aspirate, and the heavier components comprise red blood cells. The positioning part comprises the majority of the mass of the separating element and is thin so that differences in the position of the separating element with respect to the interface are small compared to differences in the densities of the separated components, particularly the component comprising red blood cells. A method allows red blood cells to move the separating element during decanting to ensure complete decant of the supernatant.
A method for separating a supernatant by centrifugation comprising providing a tubular chamber having therein a separating element, wherein said separating element is capable of floating in a physiological fluid during centrifugation, assuming a predetermined position in said physiological fluid with respect to an interface between a supernatant and a heavier component during centrifugation, and allowing air to pass said separating element to a portion of said tubular chamber having said heavier component during decanting of said supernatant, adding said physiological fluid to said tubular chamber, subjecting said tubular chamber and said physiological fluid to centrifugation, and decanting said supernatant, wherein said step of decanting includes allowing said separating element to move linearly along said tubular chamber toward said supernatant by engagement with said heavier component during said decanting.
In combination, a tubular chamber and a separating element that floats in a physiological fluid in said tubular chamber during centrifugation and assumes a predetermined position in said physiological fluid with respect to an interface between a supernatant and a heavier component, said separating element being configured to allow air to pass between an outer edge of said separating element and an inner wall of said tubular chamber to an area of said tubular chamber having said heavier component during decant of said supernatant, wherein said separating element is configured to tip in response to a force applied to one side of said separating element by said heavier component during said decanting and comprises a thin disk and a plurality of stabilizing legs for providing stability when said separating element floats within said tubular chamber.
