Abstract
Disclosed is a method for determining the presence of Mycobacterium tuberculosis complex nucleic acids in a test sample. In particular, regions of the IS6110 preferential locus (ipl) 3′-flanking region of the Mycobacterium tuberculosis complex genome are amplified and detected. In addition, oligonucleotides that can be used as primers to amplify the ipl 3′-flanking region and probe oligonucleotides are described.
Claims27
A method for detecting Mycobacterium tuberculosis complex (MT complex) nucleic acid in a sample containing nucleic acid, said method comprising:
- (a) contacting the sample with a forward and a reverse oligonucleotide primer under conditions suitable for amplification;
- (b) amplifying the sample using the primers of step (a) to generate an amplicon with mycobacterial sequence consisting of all or a portion of the IS6110 preferential locus (ipl) 3′-flanking region if present in the sample, wherein the ipl 3′-flanking region is at least 90% identical to SEQ ID NO:5;
- and
- (c) detecting said amplicons, wherein detecting indicates the presence of Mycobacterium tuberculosis complex (MT complex) nucleic acids in the sample.
A method for detecting Mycobacterium tuberculosis complex (MT complex) nucleic acid in a sample containing nucleic acid, said method comprising:
- (a) contacting the sample with a forward oligonucleotide primer comprising a sequence of at least 15 nucleotides of SEQ ID NO:
- 1 and a reverse oligonucleotide primer comprising a sequence of at least 15 nucleotides of SEQ ID NO:
- 2,
- (b) amplifying the sample using the primers of step (a) to generate all or a portion of the ipl 3′-flanking region if present in the sample to generate MT complex amplicons;
- and
- (c) detecting amplified nucleic acids, wherein detecting indicates the presence of Mycobacterium tuberculosis complex (MT complex) nucleic acids in the sample.
A method for detecting Mycobacterium tuberculosis complex (MT complex) nucleic acid in a sample containing nucleic acid, said method comprising:
- (a) contacting the sample with a forward and a reverse oligonucleotide primer under conditions suitable for amplification,
- (b) amplifying all or a portion of the IS6110 preferential locus (ipl) 3′-flanking region if present in the sample to generate MT complex amplicons, wherein the ipl 3′-flanking region is at least 90% identical to SEQ ID NO:5 and wherein said amplicons do not contain an IS6110 insertion site;
- and
- (c) detecting said amplicons, wherein detecting indicates the presence of Mycobacterium tuberculosis complex (MT complex) nucleic acids in the sample.
A method for detecting Mycobacterium tuberculosis complex (MT complex) nucleic acid in a sample containing nucleic acid, said method comprising:
- (a) contacting the sample with a forward and a reverse oligonucleotide primer under conditions suitable for amplification, wherein the forward and reverse primers hybridize to IS6110 preferential locus (ipl) 3′-flanking region;
- (b) amplifying all or a portion of the IS6110 preferential locus (ipl) 3′-flanking region if present in the sample to generate MT complex amplicons, wherein the ipl 3′-flanking region is at least 90% identical to SEQ ID NO:5;
- and
- (c) detecting said amplicons, wherein detecting indicates the presence of Mycobacterium tuberculosis complex (MT complex) nucleic acids in the sample.
