Abstract
The present invention is directed to a novel multi-spectral exogenous fluorescence polarization imaging technique that enables rapid imaging of large tissue fields. The imaging device includes a tunable monochromatic light source and a CCD camera. Linear polarizers are placed into both the incident and collected light pathways in order to obtain fluorescence polarization or/and anisotropy image. To acquire exogenous fluorescence image, fluorescent contrast agents are delivered to a target tissue.
Claims13
A method for imaging a tissue region comprising the steps of:
- applying a fluorescent contrast agent to said tissue region;
- emitting light having a first polarization directly onto the tissue region;
- detecting exogenous fluorescence light remitted from the tissue region having said first polarization direction and exogenous fluorescence light remitted from the tissue region having a second polarization direction perpendicular to said first polarization direction, wherein said remitted exogenous fluorescence light is detected from both normal and diseased tissue in the tissue region;
- forming a total exogenous fluorescence polarization image equal to said detected exogenous fluorescence light having said first polarization direction plus said detected light having said second polarization direction;
- and
- delineating margins of a lesion from superficial normal tissue in said tissue region by using a difference between the total exogenous fluorescence polarization image of the superficial normal tissue and the lesion.
A method for imaging a tissue region comprising the steps of:
- emitting light having a first polarization directly onto the tissue region;
- detecting endogenous fluorescence light remitted from the tissue region having said first polarization direction and endogenous fluorescence light remitted from the tissue region having a second polarization direction perpendicular to said first polarization direction, wherein said remitted endogenous fluorescence light is detected from both normal and diseased tissue in the tissue region;
- forming a total endogenous fluorescence polarization image based on said detected endogenous fluorescence light having said first polarization direction and said detected light having said second polarization direction
- applying a fluorescent contrast agent to said tissue region;
- emitting light having a first polarization directly onto the tissue region;
- detecting exogenous and endogenous fluorescence light remitted from the tissue region having said first polarization direction and exogenous and endogenous fluorescence light remitted from the tissue region having a second polarization direction perpendicular to said first polarization direction, wherein said remitted exogenous and endogenous fluorescence light is detected from both normal and diseased tissue in the tissue region;
- forming a total exogenous and endogenous fluorescence polarization image equal to said detected exogenous and endogenous fluorescence light having said first polarization direction plus said detected light having said second polarization direction;
- and
- subtracting the total endogenous fluorescence polarization image from the total exogenous and endogenous fluorescence polarization image to form a net exogenous fluorescence polarization image;
- and
- delineating margins of a lesion from superficial normal tissue in said tissue region by using a difference between the superficial normal tissue and the lesion in the net exogenous fluorescence polarization image.
