Abstract
Methods and materials are provided for the production of compositions of erythropoietin protein, wherein said compositions comprise a pre-selected N-linked glycosylation pattern as the predominant N-glycoform.

A method for producing an erythropoietin composition comprising the steps of:
- constructing a Pichia pastoris host cell to attach pre-selected N-linked glycans to proteins, said pre-selected N-linked glycans comprising greater than 25 mole percent of an N-linked glycan having the structure N-acetylglucosamine2Mannose3N-acetylglucosamine2Galactose2N-acetylneuraminic acid2 (GlcNAc2Man3GlcNAc2Gal2NANA2), and said host cell expressing a sequence encoding erythropoietin operably linked to expression control sequences recognizable by said host cell;
- culturing said transformed cell under conditions amenable to the expression of erythropoietin;
- and
- isolating said erythropoietin.
