Abstract
Aspects of the invention relate to methods for the production of difunctional alkanes in host cells. In particular, aspects of the invention describe components of genes associated with the difunctional alkane production from carbohydrate feedstocks in host cells. More specifically, aspects of the invention describe metabolic pathways for the production of adipic acid, aminocaproic acid, caprolactam, and hexamethylenediamine via 2-ketopimelic acid.
Claims17
A method of producing a difunctional hexane comprising:
- a) providing a recombinant host cell expressing a recombinant alpha-ketoacid decarboxylase enzyme or a recombinant 2-amino transferase enzyme capable of using alpha-ketopimelic acid as a substrate;
- b) producing alpha-ketopimelic acid in the recombinant host cell;
- and
- 1) synthesizing adipic acid semialdehyde from alpha-ketopimelic acid, wherein the recombinant alpha-keto acid decarboxylase enzyme catalyses decarboxylation of alpha-ketopimelic acid to form adipic acid semialdehyde, or
- 2) synthesizing 2-amino pimelic acid from alpha-ketopimelic acid, wherein the recombinant 2-amino transferase enzyme catalyzes conversion of alpha-ketopimelic acid to form 2-amino pimelic acid.
A method for the production of adipic acid from alpha-ketopimelic acid, comprising:
- a) providing a recombinant host cell which is capable of producing alpha-ketopimelic acid;
- b) expressing a recombinant alpha-ketoacid decarboxylase enzyme and a recombinant aldehyde dehydrogenase enzyme in the recombinant host cell;
- and
- c) synthesizing adipic acid by catalyzing the conversion of alpha-ketopimelic acid to adipic semialdehyde with a recombinant alpha-ketoacid decarboxylase and by catalyzing the conversion of adipic semialdehyde to adipic acid with a recombinant aldehyde dehydrogenase, wherein the recombinant alpha-ketoacid decarboxylase and the recombinant aldehyde dehydrogenase are expressed in the recombinant host cell, thereby to produce adipic acid.
A method for the production of amino-caproic acid from alpha-ketopimelic acid comprising:
- a) providing a recombinant host cell which is capable of producing alpha-ketopimelic acid;
- b) expressing a recombinant alpha-ketoacid decarboxylase enzyme and a recombinant aminotransferase enzyme or a recombinant 2-amino-transferase enzyme and a recombinant 2-aminodecarboxylase enzyme in the recombinant host cell;
- and
- c) synthesizing amino caproic acid by (1) catalyzing the conversion of alpha-ketopimelic acid to adipic semialdehyde with a recombinant alpha-ketoacid decarboxylase and (2) catalyzing the conversion of adipic semialdehyde to amino caproic acid with a recombinant aminotransferase enzyme, wherein the recombinant alpha-ketoacid decarboxylase and the recombinant aminotransferase enzymes are expressed in the host cell;
- or
- synthesizing amino caproic acid by (1) catalyzing the conversion of alpha-ketopimelic acid to 2-amino pimelic acid with a recombinant 2-amino-transferase and (2) catalyzing the conversion of 2-amino pimelic acid to caproic acid with a recombinant 2-aminodecarboxylase, wherein the recombinant 2-amino-transferase and the recombinant 2-aminodecarboxylase enzymes are expressed in the host cell,
- thereby to produce amino-caproic acid.
