Abstract
The invention concerns a novel method for making herbicide-tolerant plants, in particular to HPPD inhibiting herbicides, the nucleic acid sequences coding for enzymes capable of being used in said method, expression cassettes containing them and transgenic plants comprising at least one of said expression cassettes.
Claims19
A method for making plants tolerant to a herbicide, characterized in that at least one enzyme making it possible to bypass the metabolic pathway inhibited by said herbicide is expressed in said plants, wherein said at least one enzyme includes HPAH and HPAC, and wherein said HPAH is encoded by a polynucleotide selected from the group consisting of the coding sequences of SEQ ID No. 7 and SEQ ID No. 17, and polynucleotides capable of selectively hybridizing to said coding sequence of SEQ ID No. 7 or SEQ ID No. 17 under the following conditions:
- prehybridization for four hours at 68° C. in a solution of 5×SSC, 6% Denhardt's, and 0.5% SDS;
- hybridization for sixteen hours at 68° C.;
- and washes at 65° C. in 2×SSC, 0.1% SDS,
- and said HPAC is encoded by a polynucleotide selected from the group consisting of the coding sequences of SEQ ID No. 9, SEQ ID No. 11, SEQ ID No. 13 and SEQ ID No. 19, and polynucleotides capable of selectively hybridizing to said coding sequence of SEQ ID No. 9, SEQ ID No. 11, SEQ ID No. 13 or SEQ ID No. 19 under the prehybridization for four hours at 68° C. in a solution of 5×SSC, 6% Denhardt's, and 0.5% SDS;
- hybridization for sixteen hours at 68° C.;
- and washes at 65° C. in 2×SSC, 0.1% SDS.
An isolated polynucleotide, selected from the group consisting of
- a) a polynucleotide that encodes an HPAH selected from the group consisting of the coding sequences of SEQ ID No. 7 and SEQ ID No. 17, and polynucleotides capable of selectively hybridizing to said coding sequence of SEQ ID No. 7 or SEQ ID No. 17 under the following conditions:
- prehybridization for four hours at 68° C. in a solution of 5×SSC, 6% Denhardt's, and 0.5% SDS;
- hybridization for sixteen hours at 68° C.;
- and washes at 65° C. in 2×SSC, 0.1% SDS, and
- b) a polynucleotide that encodes an HPAC selected from the group consisting of the coding sequences of SEQ ID No. 9, SEQ ID No. 11, SEQ ID No. 13 and SEQ ID No. 19, and polynucleotides capable of selectively hybridizing to said coding sequence of SEQ ID No. 9, SEQ ID No. 11, SEQ ID No. 13 or SEQ ID No. 19 under the following conditions:
- prehybridization for four hours at 68° C. in a solution of 5×SSC, 6% Denhardt's, and 0.5% SDS;
- hybridization for sixteen hours at 68° C.;
- and washes at 65° C. in 2×SSC, 0.1% SDS.
