Screening methods involving the detection of short-lived proteins

A method for identifying relatively short-lived proteins, which comprises:

• providing cells, wherein the cells express fusion proteins comprising a reporter protein and a protein encoded by a sequence from a cDNA library;
• partitioning the cells into populations of cells based on an intensity of a reporter signal from the fusion protein;
• inhibiting further protein expression;
• measuring signal intensities from the reporter proteins;
• selecting a population of cells from the populations of cells based on the measured signal intensities;
• and
• identifying a relatively short-lived protein.

A method for identifying relatively short-lived proteins, which comprises:

• providing cells, wherein the cells express fusion proteins comprising a reporter protein and a protein encoded by a sequence from a cDNA library;
• partitioning the cells into populations of cells based on an intensity of a reporter signal from the fusion protein;
• inhibiting protein degradation;
• measuring signal intensities from the reporter proteins;
• and
• selecting a population of cells from the populations of cells based on the measured signal intensities, thereby identifying a relatively short-lived protein.
