Abstract
Disclosed are mutant DNA polymerases having improved extension rates relative to a corresponding, unmodified polymerase. The mutant polymerases are useful in a variety of disclosed primer extension methods. Also disclosed are related compositions, including recombinant nucleic acids, vectors, and host cells, which are useful, e.g., for production of the mutant DNA polymerases.
Claims16
A DNA polymerase wherein the polymerase has at least 90% sequence identity to SEQ ID NO:20 or SEQ ID NO:21, comprising A-G-X1-X2-F-X3-X4-X5-S-X6-X7-Q-X8-X9-X10-X11-L-X12-X13-X14-X15(SEQ ID NO:33), wherein
- X2, X5, X6, X9, and X10 are any amino acid,
- X1 is H, E or Q,
- X3 is N or H,
- X4 is L or I,
- X7 is D, K or T,
- X8 is L or V,
- X11 is V, I or L,
- X12 is F or Y,
- X13 is G,
- X14 is K or E, and
- X15 is L or Q;
- wherein the polymerase has an increased nucleic acid extension rate relative to a DNA polymerase that is identical with the exception that X13 is E.
