Abstract
A method for enhancing absorption of a compound in a medicament that is suitable for administering transmucosally to a subject is conducted in an environment having a suitable pH. The suitable pH is determined based on the ionization constant (dissociation constant) of the compound in water, the solubility of the ionized form of the compound, and the solubility of the non-ionized form of the compound.
Claims15
A method for enhancing transmucosal absorption of a medicament comprising a compound or a pharmaceutically acceptable salt thereof that is suitable for transmucosal administration to a subject, said method comprising calculating a pH according to the following equation:
- pH
- =

- wherein Ka is the ionization constant (dissociation constant) of the compound in water, Si is the solubility of the ionized form of the compound in water, and Su is the solubility of the non-ionized form of the compound in water;
- and
- adjusting an environment for the administration to be about said pH;
- by administering said medicament to the environment in a buffer composition that produces said pH when delivered to the environment.
