Newly identified mammalian taste-cell-specific G protein-coupled receptors, and the genes and cDNA encoding said receptors are described. Specifically, T1R G protein-coupled receptors active in taste signaling, and the genes and cDNA encoding the same, are described, along with methods for isolating such genes and for isolating and expressing such receptors. Methods for representing taste perception of a particular tastant in a mammal are also described, as are methods for generating novel molecules or combinations of molecules that elicit a predetermined taste perception in a mammal, and methods for simulating one or more tastes. Further, methods for stimulating or blocking taste perception in a mammal are also disclosed.
A chimeric T1R1 taste receptor polypeptide containing the transmembrane domains of the human T1R1 polypeptide in SEQ ID NO:17 and the extracellular domains of a GPCR other than the human T1R1 in SEQ ID NO:17 wherein the transmembrane domains in said chimeric T1R1 taste receptor polypeptide are at least 90% identical to the entire transmembrane domains of the human T1R1 polypeptide in SEQ ID NO:17 and the extracellular domains are at least 90% identical to the extracellular domains of another GPCR polypeptide wherein said another GPCR is endogenous (wild-type) human or rodent T1R taste receptor polypeptide other than human T1R1 or a GPCR polypeptide other than a T1R taste receptor polypeptide and wherein a taste receptor comprising said chimeric T1R1 taste receptor polypeptide specifically binds or functionally responds to ligands that specifically bind or modulate the activity of a taste receptor comprising the T1R1 polypeptide in SEQ ID NO:17.
