• X is O

• R5 is selected from H, hydroxy, amino, alkoxy, alkylthio, sulfonyl, C1-10alkyl, C2-6alkenyl, C2-6alkynyl, aryl, heteroaryl, and heterocyclyl, and
• Q is N;
• Ring b is selected from
• aryl;
• heteroaryl;
• and
• heterocyclyl of the formula
• wherein
• G1 and G2 are independently selected from N, C, and CH;
• and
• D1 and D2 are each 1-3 independent members selected from CH, CH2, N, S, and O, provided that when G1 or G2 is N, D1 and D2 are independently selected from CH and CH2;
• R1 is selected from halo, nitrile, hydroxyl, thiol, amino, alkoxy, alkylthio, sulfonyl, C1-10alkyl, C2-6alkenyl, C2-6alkynyl, cycloalkyl, aryl, heteroaryl, heterocyclyl, carbonyl, and —CHO;
• R2 is 0-2 independent members selected from halo, nitrile, hydroxyl, amino, C1-10alkyl, C2-6alkenyl, C2-6alkynyl, alkoxy, alkylthio, sulfonyl, aryl, heteroaryl, heterocyclyl, —C(O)R3, —C(O)OR3, and —C(O)NR3R4, wherein
• R3 and R4 are independently selected from H, C1-10alkyl, aryl, heteroaryl, and heterocyclyl, or
• R3 and R4 together with the N they are attached to form a 3-, 4-, 5-, 6-, or 7-membered heterocyclyl;
• W is selected from a covalent bond, —(CH2)p—O—, —O—(CH2)p—, —S(O)p—, —C(O)—, C1-3alkylene, C2-3alkenylene, C2-3alkynylene, and 5-7 membered aliphatic ring containing one or two nitrogens, wherein
• p is 0, 1, or 2;
• Y is selected from O, S, S(O), S(O)2, —SO2N(R6)—, —N(R6) SO2—, —N(R6) SO2N(R7)—, —N(R6)CO—, —N(R6)PO(OR8)—, —N(SO2R8)—, —N(COR8)—, —N(POOR8R9)—, —CH(OH)—,

• R6 and R7 are independently selected from H, C1-10alkylsulfonyl, arylsulfonyl, alkylcarbonyl, and arylcarbonyl, and
• R8 and R9 are independently selected from C1-6alkyl, aryl, heteroaryl, and heterocyclyl;
• Z is —CH(R10)— or —CH(R10)CH(R11)—, wherein
• R10 and R11 are independently selected from H, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, aryl, heteroaryl, and heterocyclyl;
• m is 0, 1, or 2;
• and
• n is 0 or 1 with the proviso that when n is 0, E is not N and Y is not O;
• or an optical isomer, enantiomer, diastereomer, racemate, prodrug or pharmaceutically acceptable salt thereof.
