Abstract
A gene transfer vector is prepared by introducing an exogenous gene into an inactivated virus envelope, through a freezing and thawing treatment or mixing with a detergent. There are also provided a pharmaceutical composition for gene therapy containing this gene transfer vector, a kit containing this gene transfer vector, and a gene transfer method employing this gene transfer vector.
Claims17
A method of introducing an exogenous gene into animal tissue in vivo, comprising:
- (a) providing an encapsulated gene transfer vector comprising a non-liposomal viral envelope from inactivated virus articles having F and HN membrane proteins and an exogenous gene;
- (b) subjecting the encapsulated gene transfer vector to at least five cycles of freezing then thawing;
- and
- (c) introducing into the animal tissue the encapsulated gene transfer vector to achieve introduction of the exogenous gene into the animal tissue.
An encapsulated gene transfer vector for introducing an exogenous gene into animal tissue in vivo, comprising a non-liposomal viral envelope from inactivated virus particles having F and HN membrane proteins and an exogenous gene,
- wherein the non-liposomal viral envelope is selected from a Sendai virus envelope;
- and
- wherein the encapsulated gene transfer vector has been subjected to at least five cycles of freezing then thawing.
