Abstract
Macrocyclic peptides are disclosed having the general formula: wherein R′, R3, R3′, R4, R6, X, Q, and W are described. Compositions comprising the compounds and methods for using the compounds to inhibit HCV are also disclosed.
Claims19
A compound of formula (I)
- or a pharmaceutically acceptable salt thereof, wherein
- (a) R4 is C(O)—R5, C(O)—N(R5)2, or C(O)—OR5;
- wherein each R5 is independently C1-9 alkyl, heterocylclyl, or C3-7 cycloalkyl optionally substituted with alkoxy, halo, or haloalkoxy;
- (b) R6 is H;
- (c) R3 and R3′ are each independently H or methyl;
- (d) Q is a C5-7 saturated or unsaturated chain, optionally containing one to three heteroatoms independently selected from O or S(O)m;
- wherein m is 0, 1 or 2;
- (e) W is —NH—SO2—R2;
- wherein R2 is cyclopropyl or cyclobutyl optionally substituted with C1-4 alkyl or C7-16 alkylaryl;
- (f) X is O, S, SO2, OCH2, CH2O or NH;
- (g) R′ is Het, C6-10 aryl or C7-14 alkylaryl, each optionally substituted with one or more Ra groups;
- or R′ is C3-9 cycloalkyl or C1-7 alkyl, each optionally substituted with halo, cyano, alkoxy, dialkylamino;
- provided that when X is O, then R′ is other than
- wherein Ra′ is H or Ra;
- Rx is selected from H, halo, C1-6 alkyl, C1-6 alkoxy, C3-6 cycloalkyloxy, aryl, and Het;
- Ry is selected from H, halo, CF3, C1-6 alkoxy, and C3-6 cycloalkyloxy;
- X is selected from CH and N;
- and Y and Y1 are each independently selected from CH, O, S, NH, and NRa;
- Y and Y1 are each independently selected from CH, O, S, NH, and NRa;
- and z is 0, 1, 2, or 3;
- and
- (h) each Ra is C1-6 alkyl, C1-6 alkoxy, halo, NO2, C6-10 aryl, and C7-14 alkylaryl.

A compound of formula (II)
- or a pharmaceutically acceptable salt thereof, wherein
- (a) R4 is —C(O)—NHR5 or C(O)—OR5;
- wherein R5 is C1-9 alkyl;
- (b) Q is a C5-7 saturated or unsaturated chain optionally containing one heteroatom selected from O and S(O)m;
- wherein m is 0, 1 or 2;
- (c) W is NH—SO2—R2, wherein R2 is cyclopropyl or cyclobutyl optionally substituted with C1-4 alkyl or C7-16 alkylaryl;
- (d) X is O, S, OCH2, CH2O or NH;
- (e) R′ is Het, C6-10 aryl or C7-14 alkylaryl, each optionally substituted with one or more Ra groups;
- or R′ is C3-9 cycloalkyl or C1-7 alkyl, each optionally substituted with halo, cyano, alkoxy, dialkylamino;
- provided that when X is O, then R′ is other than
- wherein Ra′ is H or Ra;
- Rx is selected from H, halo, C1-6 alkyl, C1-6 alkoxy, C3-6 cycloalkyloxy, aryl, and Het;
- Ry is selected from H, halo, CF3, C1-6 alkoxy, and C3-6 cycloalkyloxy;
- X is selected from CH and N;
- Y and Y1 are each independently selected from CH, O, S, NH, and NRa;
- and z is 0, 1, 2, or 3;
- and
- (f) each Ra is C1-6 alkyl, C1-6 alkoxy, halo, C6-10 aryl, or C7-14 alkylaryl.
