Abstract
The present invention is directed to methods of switching a differentiation of a cell from a non-osteogenic lineage into an osteogenic lineage. The present invention is also directed to methods of generating a model system for assessing the intracellular signaling pathways of bone growth factors.
Claims17
A method of switching a differentiation of a peripheral blood leukocyte from a non-osteogenic lineage into an osteogenic lineage comprising introducing into said peripheral blood leukocyte an agent capable of disrupting binding between a Smurf1 protein and a Smad 1/5 protein, wherein the agent capable of disrupting binding between Smurf-1 and Smad 1/5 is selected from i) an amino acid comprising SEQ ID NO:51;
- or ii) a nucleic acid encoding the amino acid sequence comprising SEQ ID NO:51.
A method of treating a bone void comprising:
- (a) obtaining at least one peripheral blood leukocyte;
- (b) introducing into the at least one peripheral blood leukocyte an agent capable of disrupting binding between a Smurf1 protein and a Smad1/5 protein;
- (c ) culturing the at least one peripheral blood leukocyte for a time sufficient to direct the at least one cell to osteogenic lineage;
- (d) introducing the at least one peripheral blood leukocyte into a patient having the bone void, wherein the agent capable of disrupting binding between Smurf-1 and Smad1/5 is selected from:
- i) an amino acid sequence comprising SEQ ID NO:
- 51;
- or
- ii) a nucleic acid sequence encoding the amino acid sequence comprising SEQ ID NO:
- 51.
A method of switching a differentiation of a peripheral blood leukocyte from a non-osteogenic lineage into an osteogenic lineage comprising introducing into said leukocyte an agent capable of disrupting binding between a Smurf 1 protein and a Smad 1/5 protein, wherein the agent capable of disrupting binding between Smurf-1 and Smad 1/5 comprises LMP-1.
A method of treating a bone void comprising:
- (a) obtaining at least one peripheral blood leukocyte;
- (b) introducing into the at least peripheral blood leukocyte an agent capable of disrupting binding between a Smurf1 protein and a Smad1/5 protein;
- (c ) culturing the at least one peripheral blood leukocyte for a time sufficient to direct the at least one peripheral blood leukocyte to osteogenic lineage;
- (d) introducing the at least one peripheral blood leukocyte into a patient having the bone void, wherein the agent capable of disrupting binding between Smurf-1 and Smad 1/5 comprises LMP-1.
