Abstract
The invention relates to compositions containing a polynucleotide encoding for a reporter gene, a selectable marker and a regulatory element, that provide a method for imaging cells in vivo.
Claims7
A transgenic mouse whose genome comprises a polynucleotide comprising:
- (a) a first nucleic acid encoding a light-generating gene product operably linked to a cis-acting angiopoietin-2 (Ang-2) regulatory element, wherein said cis-acting Ang-2 regulatory element comprises the nucleic acid sequence set forth in SEQ ID NO:
- 1;
- and
- (b) a second nucleic acid encoding a selectable marker, wherein said selectable marker is operably linked to a promoter, and wherein expression of said selectable marker allows for selection of said polynucleotide in a eukaryotic host.
A transgenic mouse comprising a recombinant nucleic acid molecule stably integrated into the genome of said mouse, said recombinant nucleic acid molecule comprising a cis-acting Ang-2 regulatory sequence comprising the nucleic acid sequence set forth in SEQ ID NO:
- 1 operably linked to a nucleic acid encoding a light-generating gene product.
A method for the production of a transgenic mouse, comprising
- introducing a recombinant nucleic acid molecule comprising a cis-acting Ang-2 regulatory sequence comprising the nucleic acid sequence set forth in SEQ ID NO:
- 1 operably linked to a nucleic acid encoding a light-generating gene product into a developing mouse embryo, wherein said recombinant nucleic acid molecule integrates into a germ line of said developing mouse embryo and allowing said mouse embryo to develop into a mouse capable of expressing said recombinant nucleic acid molecule.
