Abstract
The invention relates to modified neomycin phosphotransferase genes and their use in a selection method for high-producing recombinant cells. The invention further relates to expression vectors which contain a modified neomycin phosphotransferase gene and a gene of interest functionally linked to a heterologous promoter and a method of preparing heterologous gene products using these expression vectors.
Claims31
A modified neomycin phosphotransferase gene wherein the modified neomycin phosphotransferase gene encodes a polypeptide comprising the amino acid sequence of SEQ ID NO:2 except that the amino acid at position 182 and/or 227 and/or 261 is a different amino acid than that of SEQ ID NO:2, wherein a glycine or an aspartic acid is at amino acid position 182 and/or an alanine or a valine or a glycine is at amino acid position 227 and/or a glycine or an asparagine is at position 261.
