Abstract
Disclosed herein are analogs of clozapine and pharmaceutically acceptable salts, esters, amides, or prodrugs thereof, methods of synthesizing the analogs; and methods of using the analogs for treating neuropsychiatric disorders. In some embodiments, the analogs are amino substituted diaryl[a,d]cycloheptenes.
Claims26
A method of treating cognitive impairment associated with Alzheimer's disease, schizophrenia or related idiopathic psychoses, Huntington's disease, Tourette's syndrome, anxiety, affective disorders, drug-induced psychoses, psychoses secondary to neurodegenerative disorders, major depression, bipolar disorder, depression with psychotic features, or sleep disorders, comprising administering to a subject exhibiting one or more symptoms of cognitive impairment a therapeutically effective amount of a compound of Formula I:
- or a pharmaceutically acceptable salt thereof, wherein:
- A has the structure:
- wherein each bond represented by a dashed and solid line in A represents a carbon-carbon single bond;
- X is nitrogen;
- X′ is C;
- Y is nitrogen;
- W is nitrogen;
- each n is 1;
- R1 is hydrogen;
- L is absent;
- a, b, c, and d are each carbon,
- e, f, g, and h are each carbon,
- R2, R3, R4, and R5, are each separately selected from the group consisting of hydrogen, halogen, optionally substituted C1-6 alkyl, optionally substituted C1-6 alkyloxy, optionally substituted C2-6 alkenyl, optionally substituted C2-6 alkynyl, optionally substituted C1-6-alkoxyalkyl, optionally substituted C1-6 alkylthio, perhaloalkyl, CN, COR10, CONHR10, NHCONHR10, SO2NHR10, SO2R10, OSO2R10, NO2, NHCOR10,
- or R2 and R3, or R3 and R4, or R4 and R5 taken together, along with the ring carbons to which they are attached, form a five-membered or six-membered cycloalkyl, heterocyclyl or heteroaryl ring, or a six-membered aryl ring moiety;
- R6, R7, R8, and R9, are each separately selected from the group consisting of hydrogen, halogen, optionally substituted C1-6 alkyl, optionally substituted C1-6 alkyloxy, optionally substituted C2-6 alkenyl, optionally substituted C2-6 alkynyl, optionally substituted C1-6-alkoxyalkyl, optionally substituted C1-6 alkylthio, perhaloalkyl, CN, COR10, CONHR10, NHCONHR10, SO2NHR10, SO2R10, OSO2R10, NO2, NHCOR10,
- or R6 and R7, or R7 and R8, or R8 and R9 taken together, along with the ring carbons to which they are attached, form a five-membered or six-membered cycloalkyl, heterocyclyl or heteroaryl ring, or a six-membered aryl ring moiety;
- Z is oxygen;
- R10 is selected from the group consisting of hydrogen, optionally substituted C1-6 alkyl, optionally substituted C3-8 cycloalkyl, optionally substituted C2-6 alkenyl, optionally substituted C2-6 alkynyl optionally substituted aryl, optionally substituted arylalkyl, and perhaloalkyl;
