Abstract
The present disclosure relates to vectors comprising polynucleotide sequences that encode HIV polypeptides. In particular, the disclosure relates polycistronic vector constructs comprising sequences that encode HIV polypeptides as a single polyprotein. Compositions comprising these vectors and sequences along with methods of using these vectors and sequences are also disclosed.
Claims32
A vector comprising a synthetic polynucleotide comprising polynucleotide sequences encoding a polyprotein comprising HIV Gag, Pol, Tat, Rev and Nef polypeptides, wherein the polynucleotide sequence is operably linked to control elements, wherein the polynucleotide sequences encoding Gag and Pol polypeptides are located 3′ to polynucleotide sequences encoding the Tat, Rev, and Nef polypeptides, wherein expression of the polynucleotide sequence is increased relative to expression of a native polynucleotide sequence encoding the polypeptides.
