Abstract
This invention provides compositions and methods for producing translational components that expand the number of genetically encoded amino acids in eukaryotic cells. The components include orthogonal tRNAs, orthogonal aminoacyl-tRNA synthetases, orthogonal pairs of tRNAs/synthetases and unnatural amino acids. Proteins and methods of producing proteins with unnatural amino acids in eukaryotic cells are also provided.
Claims10
A kit for producing a protein that comprises at least one unnatural amino acid in a eukaryotic cell, the kit comprising:
- a container containing a polynucleotide sequence encoding an orthogonal tRNA (O-tRNA), and a polynucleotide sequence encoding an orthogonal aminoacyl tRNA synthetase (O-RS),
- wherein:
- the O-RS preferentially aminoacylates the O-tRNA with the unnatural amino acid, and wherein the O-RS comprises the polypeptide sequence selected from the group consisting of:
- SEQ ID NO:
- 36-47 and 86;
- the cell produces the protein in the absence of the unnatural amino acid with a yield that is less than 10% of the yield in the presence of the unnatural amino acid;
- and the O-tRNA specifically recognizes a selector codon in a recombinant nucleic acid of interest in the cell.
