Abstract
Tumor necrosis factor receptor DNAs and expression vectors encoding TNF receptors, and processes for producing TNF receptors as products of recombinant cell culture, are disclosed.
Claims30
An isolated glycoprotein that is capable of binding TNF, wherein said glycoprotein comprises amino acids 1-163 of SEQ ID NO:2 and is not isolated from human urine.
An isolated mammalian cell which can be propagated in vitro and which, upon growth in culture medium, produces a glycoprotein comprising amino acids 1-163 of SEQ ID NO:2, wherein said glycoprotein is capable of binding TNF, and wherein said cell comprises non-human DNA sequences that control transcription of DNA encoding said glycoprotein.
A composition comprising a the glycoprotein comprising amino acids 1-163 of SEQ ID NO:2 and a pharmaceutically acceptable diluent or carrier, wherein said glycoprotein is purified from culture medium of mammalian cells grown in culture, and wherein said glycoprotein is capable of binding TNF.
An assay for detecting TNF comprising contacting a test sample comprising TNF with an isolated and purified biologically active mammalian TNF receptor comprising amino acids 1-163 of SEQ ID NO:2 and detecting binding between said TNF and said TNF receptor.
An assay for detecting TNFR molecules comprising contacting a test sample comprising TNFR molecules and TNF with an isolated and purified biologically active mammalian TNF receptor comprising amino acids 1-163 of SEQ ID NO:2 and detecting competitive binding between said TNF and said TNF receptor and said TNFR molecules.
An assay for detecting the interaction between TNF and TNFR molecules comprising contacting a test sample comprising TNF with an isolated and purified biologically active mammalian TNF receptor comprising amino acids 1-163 of SEQ ID NO:2 which has been labeled and detecting binding between said TNF and said labeled TNFR.
