Abstract
The present invention provides methods to produce immunotoxins (ITs) and cytokines with a reduced ability to promote vascular leak syndrome (VLS). The invention also provides ITs and cytokines which have been mutated to lack amino acid sequences which induce VLS.
Claims22
An immunotoxin comprising at least one antibody or an antigen binding fragment thereof and a modified ricin A chain toxin having an LDV sequence, wherein the modified toxin comprises at least one amino acid mutation in R48 or N97 as set forth in SEQ ID NO:
- 1 that reduces the ability of the toxin to induce vascular leak syndrome relative to the native toxin having the amino acid sequence as set forth in SEQ ID NO:
- 1.
An immunotoxin comprising at least one antibody or an antigen binding fragment thereof and a modified ricin A chain toxin having an LDV sequence, wherein the modified toxin comprises at least one amino acid mutation in L74, D75, or V76 as set forth in SEQ ID NO:
- 1 that reduces the ability of the toxin to induce vascular leak syndrome relative to the native toxin having the amino acid sequence as set forth in SEQ ID NO:
- 1.
