Abstract
This document provides methods and materials related to rapid, quantitative determination of TPMT activity in biological samples. Also featured are compositions and kits useful for determination of TPMT activity in biological samples.
Claims12
A method for determining the level of a product of thiopurine methyltransferase (TPMT) activity in a biological sample, comprising:
- a) contacting said biological sample with a TPMT substrate, a methyl donor, and an internal standard consisting of isotopically-labeled methylated TPMT substrate to form a reactant mixture;
- b) extracting said mixture with organic solvent to form an organic extracted fraction comprising said internal standard and, if present, enzymatically produced methylated TPMT substrate;
- and
- c) measuring levels of said internal standard and said methylated TPMT substrate in said extracted fraction using liquid chromatography tandem mass spectrometry (LC-MS/MS), wherein said internal standard and said methylated TPMT substrate elute together.
