Abstract
Conjugates of Factor VII (FVII) and Factor VIIa (FVIIA) are provided, as are methods for preparing them. Methods for producing novel polypeptides contributing to the production of such conjugates are provided. Methods of treatment by administering a FVII or FVIIa conjugate are provided.
Claims27
A method for producing a conjugate, said conjugate comprising (i) a polypeptide having clotting activity, said polypeptide comprising an amino acid sequence which (a) differs from the human Factor (hFVII) or human Factor VIIa (hFVIIa) sequence shown in SEQ ID NO:1 in no more than 15 amino acid residues and (b) comprises an introduced N-glycosylation site relative to SEQ ID NO:1, wherein the introduced N-glycosylation site comprises the substitution V253N, wherein amino acid residue positions in said polypeptide are numbered according to SEQ ID NO:1:
- and (ii) a sugar moiety covalently attached to the introduced N-glycosylation site, the method comprising:
- culturing a host cell comprising an expression vector comprising a nucleotide sequence which encodes said polypeptide under conditions conducive for the expression of the polypeptide, and recovering the polypeptide, wherein (a) the host cell is a eukaryotic host cell capable of in vivo glycosylation, and/or (b) the polypeptide is subjected to conjugation to a sugar moiety in vitro.
