## Abstract

A method for improving the diagnostic assessment of cartilage degenerative processes, and to provide means of monitoring the effects of therapeutical measures taken towards arthritic diseases in most mammals utilizes an immunoassay to detect fragments of collagen type II resulting from collagenase activity comprising an antibody directed against an epitope comprised in the amino acid sequence HRGYPGLDG, located in the helical region of collagen type II.

## Claims

### Claim 1

A method of qualitative or quantitative assay of unwound collagen type II fragments containing amino acid sequence HRGYPGLDG (SEQ ID NO: 1) in a biological sample comprising
contacting the fragments with an antibody that binds with an epitope in the amino acid sequence HRGYPGLDG (SEQ ID NO: 1) and
detecting resulting immunoreaction.

### Claim 2 (depends on 1)

The method of claim 1, wherein the epitope consists of five or more amino acids in the sequence.

### Claim 3 (depends on 1)

The method of claim 1, wherein the antibody does not react with wound collagen type II fragments.

### Claim 4 (depends on 1)

The method of claim 1, wherein immunoreaction is detected between the epitope and collagen type II fragments of less than 80 kDa.

### Claim 5 (depends on 4)

The method of claim 4, wherein the fragments are of less than 30 kDa.

### Claim 6 (depends on 4)

The method of claim 4, wherein the fragments are of less than 10 kDa.

### Claim 7 (depends on 1)

The method of claim 1, wherein the sample is a biological fluid.

### Claim 8 (depends on 7)

The method of claim 7, wherein the biological fluid is serum, urine, tear fluid, synovial fluid, subcutaneous interstitial fluid, a cell culture supernatant fluid, or a cell extract.

### Claim 9 (depends on 1)

The method of claim 1, wherein detected immunoreaction is indicative of degradation of cartilage.
